Enablex Information:

Enablex (Darifenacin) is an antimuscarinic agent used to treat symptoms of overactive bladder such as sudden and intense urges to urinate that are difficult to delay, abnormally high urinary frequency during the day or night, and urge incontinence, which involves accidental urine leakage immediately following a strong urge.

This medication works by blocking specific chemical signals to relax the bladder muscle. Normally, the body releases a chemical messenger called acetylcholine, which binds to M3 muscarinic receptors on the bladder's smooth muscle. This attachment triggers the muscle to contract, pushing urine out. In an overactive bladder, these contractions happen too early or too often, creating sudden, intense urges to urinate.

Darifenacin attaches itself directly to these M3 receptors, physically blocking acetylcholine from binding to them. By interrupting this chemical signal, the medication reduces involuntary bladder contractions. This allows the bladder muscle to remain relaxed, increasing the amount of urine it can comfortably hold and significantly delaying the urge to urinate. Extended-release tablets must be swallowed whole and should not be chewed, crushed, or divided.

Enablex Side Effects

Like other medications in its drug class, darifenacin is associated with a range of side effects, mostly associated with its antimuscarinic properties. Some common mild side effects include constipation, dry mouth, headache, heartburn, nausea, urinary tract infection, blurred vision, dizziness, and tiredness. Other possible side effects include diarrhea, runny or stuffy nose, trouble sleeping, changed sense of taste, and cough. Kindly inform your doctor if you experience any of these side effects, and if they persist or worsen.

Stop taking this medication and seek immediate medical attention if you notice the following while taking this medication:
• Allergic reactions such as breathing difficulties, body rash, itching, hives, skin blistering, swelling of the face, lips, or throat;
• Sudden inability to urinate, with lower abdominal pain (urinary retention);

Other possible serious side effects of this medication include:
• Facial swelling;
• Increased blood pressure;
• Signs of a urinary tract infection (e.g., difficult or painful urination, or blood in the urine);
• Swelling of the feet, ankles, or lower leg;
• Persistent vision changes;

Enablex Precautions

Do not use this medication if you have:
• Allergic reaction to darifenacin, or any other ingredients in Enablex;
• Delayed emptying of the stomach (gastric retention);
• Difficulty urinating or emptying the bladder (urinary retention);
• Uncontrolled narrow-angle glaucoma;

This medication needs to be used with caution in some individuals with certain medical conditions. Before starting this medication, ensure that you inform your doctor about any prior or current medical conditions, including the following:
• Diabetes;
• Eating disorders;
• Electrolyte imbalances;
• Gastrointestinal disorders such as intestinal blockage and ulcerative colitis;
• Heart disease or high blood pressure;
• Kidney disease;
• Liver disease (moderate liver impairment requires a strict maximum dose limit of 7.5 mg daily, and Enablex is not recommended for individuals with severe liver disease);
• Muscle problems such as myasthenia gravis;
• Nerve problems such as autonomic neuropathy;
• Severe constipation or severe gastrointestinal motility disorders;
• Stroke;

Enablex can cause blurred vision, mild drowsiness, and dizziness. It is advised to exercise caution when driving, operating heavy machinery, or performing dangerous tasks until you fully understand how the medication affects your alertness and vision. Taking darifenacin with certain medications, such as antifungals (like ketoconazole or itraconazole) or antibiotics (like erythromycin and clarithromycin), usually requires careful dosage adjustments, as these medications can alter its liver metabolism. Therefore, before using this medication, inform your doctor about any other medications you are currently taking, including herbal medicines and supplements, to determine if it is safe to use along with darifenacin.

There are no adequate, well-controlled clinical trials evaluating darifenacin use in pregnant women, so it should only be used if the benefit clearly outweighs the potential risk to the fetus. It also remains unknown whether this medication passes into human breast milk. Animal data indicate that this medication is excreted into milk; as a result, healthcare providers advise exercising caution when breastfeeding. Additionally, its anticholinergic properties could potentially suppress a mother's overall breast milk supply. This medication is not suitable for use in children. Avoid exposure to hot environments or strenuous physical exertion, as this medication can reduce sweating, increasing the risk of severe heat exhaustion and heat stroke. Store this medication at room temperature, protect it from light and moisture, and keep it out of the reach of children.
