Ferriprox (Deferiprone) Information

Ferriprox, which contains the active ingredient deferiprone, is a prescription iron chelator used to treat transfusional iron overload in patients with thalassemia syndromes, sickle cell disease, or other anemias. It is typically reserved for patients who have had an inadequate response to other chelation therapies, such as deferoxamine.

The mechanism of action involves acting as an iron chelator (binder). It enters the bloodstream and binds specifically to ferric iron, forming a stable 3:1 complex. This complex is then excreted by the kidneys into the urine, effectively removing excess iron from the body's tissues and organs. As a result of this process, your urine may turn reddish-brown while taking this medication.

Ferriprox Side Effects

Ferriprox is generally well-tolerated, but common side effects may occur as your body adjusts to the medication. Commonly reported effects include gastrointestinal distress such as nausea, vomiting, abdominal pain, and diarrhea. Many people also experience joint pain (arthralgia) or stiffness, as well as an increased appetite, headache, and changes in liver function tests. To minimize stomach upset when first starting the drug, your doctor may recommend a lower initial dose that is gradually increased over several weeks.

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Drop in white blood cell count (agranulocytosis). This is life-threatening, and your doctor will recommend weekly blood tests to monitor for agranulocytosis.
• Signs of an infection, including fever, chills, sore throat, flu-like symptoms, or mouth sores

Ferriprox Precautions

Do not use Ferriprox if you have a known allergy to deferiprone or any inactive ingredient in the medication. Before starting treatment, inform your doctor of your complete medical history, especially if you have a history of neutropenia or agranulocytosis, liver disease, kidney impairment, or zinc deficiency. Your liver enzymes should be monitored monthly, and your zinc levels should be checked annually or if you develop symptoms of zinc deficiency.

Tell your doctor about all medications you are taking, including prescription drugs, over-the-counter medicines, vitamins, and herbal supplements. Ferriprox may interact with drugs or supplements containing polyvalent cations, such as iron, aluminum, zinc, or calcium. You should allow at least a 4-hour interval between taking Ferriprox and these products. Avoid using other iron chelators, such as deferasirox or deferoxamine, unless specifically instructed by your doctor.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Ferriprox is strictly contraindicated during pregnancy as it can cause fetal harm, including congenital disabilities or pregnancy loss. Both men and women should use effective birth control during treatment. Women should continue birth control for at least 6 months after the last dose. Breastfeeding is not recommended while taking this medication and for at least 2 weeks after the final dose, as it is unknown if the drug passes into breast milk. Use Ferriprox exactly as your doctor tells you. Your dosage will depend on the formulation prescribed. To reduce the risk of nausea, it is recommended to take the medication with food. If you are using the oral solution, always use the calibrated measuring cup provided with the bottle to ensure an accurate dose.

Do not take alcohol when using this medicine. Store the tablets or oral solution at room temperature in the original container, away from heat, moisture, and direct light. Keep the bottle tightly closed when not in use. For the oral solution, any remaining liquid must be discarded 35 days after opening the bottle. Keep out of reach of children.
