Lynkuet (Elinzanetant) Information

Lynkuet, which contains the active ingredient elinzanetant, is a non-hormonal prescription medication used to treat moderate to severe vasomotor symptoms (hot flashes and night sweats) associated with menopause. It is also indicated for hot flashes caused by adjuvant endocrine therapy for breast cancer. Approved in 2025, it is the first and only dual neurokinin 1 (NK1) and neurokinin 3 (NK3) receptor antagonist.

The mechanism of action involves targeting specific KNDy neurons in the hypothalamus, the brain's temperature control center. During menopause or endocrine therapy, declining estrogen levels cause these neurons to become overactive and release excess neurokinin B, which disrupts thermoregulation. Elinzanetant blocks neurokinin B from binding to NK1 and NK3 receptors, helping restore normal body temperature regulation, reducing the frequency and intensity of hot flashes, and improving sleep quality.

Lynkuet Side Effects

Lynkuet is generally well-tolerated, but common side effects may occur as your body adjusts to the medication. Commonly reported effects include headache, fatigue, dizziness, and somnolence (feeling drowsy or sleepy). Some individuals may also experience gastrointestinal issues such as abdominal pain, diarrhea, nausea, or gastroesophageal reflux disease. Other reported effects include a skin rash, muscle spasms, and nasopharyngitis (cold-like symptoms).

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Hepatotoxicity (Liver Injury): Signs include jaundice (yellowing of the eyes or skin), dark urine, pale stools, severe upper right stomach pain, or unusual fatigue.
• Seizures: Especially in individuals with a history of seizure disorders.
• Severe Allergic Reactions: Swelling of the face, lips, or throat, hives, or significant difficulty breathing.
• Severe CNS Depression: Extreme confusion or an inability to wake up.

Lynkuet Precautions

Do not use Lynkuet if you have a known allergy to elinzanetant or any inactive ingredient in the medication. Before starting treatment, inform your doctor of your complete medical history, especially if you have liver or gallbladder problems, a history of seizures, or kidney disease.

Tell your doctor about all medications you are taking, including prescription drugs, over-the-counter medicines, vitamins, and herbal supplements. Lynkuet has significant interactions with many drugs, especially strong CYP3A inhibitors like clarithromycin and ketoconazole. Do not consume grapefruit or its juice, as it significantly increases Lynkuet levels in the blood.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Lynkuet is strictly contraindicated in pregnancy. Animal studies suggest the medication can cause pregnancy loss or stillbirth. Females of reproductive potential must have a pregnancy test before starting treatment. They should use effective birth control during therapy and for at least 2 weeks after the last dose. It is unknown if elinzanetant passes into human breast milk. Consult your doctor before breastfeeding while using this medication.

Take Lynkuet exactly as your doctor tells you. Swallow the capsules whole with water at approximately the same time each day, preferably at bedtime due to the potential for drowsiness. Do not cut, crush, or chew the capsules, as they contain an oily solution. Because the medication can cause dizziness and daytime impairment, avoid driving or engaging in hazardous activities until you know how the drug affects you. Store Lynkuet at room temperature (68°F to 77°F) in its original packaging to protect it from moisture and light. Keep them out of reach of children.
