Sinequan Information

Sinequan (Doxepin) is used to treat major depression. Depression is a mental health condition characterized by persistent sadness, loss of interest, fatigue, and changes in sleep or appetite that can interfere with daily functioning. Sinequan is typically used in individuals with depression who also have anxiety. It belongs to the older class of antidepressants called tricyclic antidepressants.

While its mechanism of act is not fully understood, it is believed to act as a serotonin-norepinephrine reuptake inhibitor, essentially increasing the levels of the chemical messengers norepinephrine and serotonin in the brain by preventing them from being reabsorbed too quickly. Higher levels of these neurotransmitters appear to help improve mood and reduce anxiety.

In addition to its effects as a serotonin-norepinephrine reuptake inhibitor, doxepin is a very potent blocker of histamine H1 receptors. Blocking this receptor in the brain causes significant sedation, which is why doxepin is useful in the treatment of insomnia, even in low doses. This same action also helps suppress allergic reactions, such as chronic hives. Doxepin may also be used to treat people with alcoholism who also have anxiety or depression.

Sinequan Side Effects

Like any other medication, doxepin can cause a range of side effects, some of which are more common and others that are serious but rare. Some common side effects of this medication include constipation, dizziness, dry mouth, drowsiness, headache, nausea, and weight gain.

While serious side effects are rare, they can occur, especially in the case of an overdose or drug interactions. Therefore, if you experience any of the following, you should stop using the medication and seek immediate medical attention:
• Allergic reactions with symptoms such as widespread rash, hives, severe itching, or swelling of the face, lips, and tongue, along with difficulty breathing;
• Cardiovascular problems such as fast, pounding, or irregular heartbeat, or fainting spells;
• Central nervous system effects such as confusion, disorientation, agitation, hallucinations, or seizures;
• Serotonin Syndrome with symptoms such as symptoms like fever, seizures, muscle spasms or twitches, nausea, and vomiting;
• Urinating difficulties

Like many antidepressants, there is a risk of increased suicidal thoughts and behavior, especially in children, adolescents, and young adults (under 25), particularly when starting the medication or changing the dose. It is essential to monitor individuals taking this medication, particularly children and young adults, for any unusual changes in behavior.

Sinequan Precautions

Do not use this oral medication if you are allergic to doxepin, other ingredients in Sinequan, or other tricyclic antidepressants. Before you start taking this medication, ensure that you inform your doctor about any prior or current medical conditions, including the following:
• Blood disorders;
• Bipolar disorder/mania;
• Diabetes;
• Eye conditions such as glaucoma;
• Heart Disease;
• High blood pressure;
• Liver disease;
• Seizure disorders;
• Suicidal ideation and thoughts;
• Thyroid disease;
• Urinating difficulties

Abrupt stoppage of this medication can cause severe withdrawal symptoms. Therefore, consult your doctor if you wish to discontinue this medication. Please don't drive or operate heavy machinery while taking this medication until you know how it affects you, as it may cause drowsiness and sedation. Avoid the intake of alcohol as it can significantly increase sedation, worsen symptoms of depression. If you have a planned surgery, talk to your doctor about your medication use. You may be asked to stop taking this medication.

Do not use this medication during pregnancy or breastfeeding without consulting your doctor. Taking doxepin with, or within two weeks of stopping a monoamine oxidase inhibitor (such as tranylcypromine and phenelzine) may cause a severe reaction called serotonin syndrome. Therefore, inform your healthcare provider about all the medicines, including herbal medications, that you are taking before using Sinequan to prevent potential drug interactions.

Take Sinequan capsules orally as directed by your doctor or as indicated on the medication label. Take it with or without food, but use food if it upsets your stomach. Do not stop suddenly; your doctor will guide you on how to stop safely. Store this medication at room temperature, protect it from light and moisture, and keep it out of the reach of children.
