Cabometyx Information
Cabometyx (Cabozantinib) is a targeted chemotherapy used alone or in combination with nivolumab as first-line treatment of individuals with renal carcinoma or for individuals with this condition who have previously received anti-angiogenic therapy. It is also used in the treatment of differentiated thyroid cancer that has become resistant to radioactive iodine therapy and has progressed after prior therapy. It may also be used to treat medullary thyroid cancer, especially metastatic disease. It is also used in the treatment of hepatocellular carcinoma, specifically for individuals who have already been treated with sorafenib. Furthermore, it may also be used to treat neuroendocrine tumours affecting the pancreas and other organs that have previously been treated but with a poor response.
Its active ingredient is cabozantinib, a potent tyrosine kinase inhibitor. This medication works as a multi-kinase antagonist, specifically inhibiting different pathways. In the context of the aggressive malignancies listed above, the disease often progresses by hijacking the body’s signaling pathways to promote the growth of new blood vessels (angiogenesis) and to invade surrounding tissues. Cabozantinib works by basically starving the tumor of its blood supply through vascular endothelial growth factor inhibition, while simultaneously blocking the MET and AXL receptors that typically allow cancer cells to develop resistance and metastasize. By disrupting these essential survival signals, the medication appears to effectively slow or shrink tumor growth and prolong progression-free survival in patients whose cancers have otherwise become difficult to manage with standard interventions.
Cabometyx Side Effects
Like all medications, Cabometyx can cause some side effects. Some mild side effects of this medication include dizziness, diarrhea, nausea, vomiting, mouth sores, constipation, stomach pain, weight loss, decreased appetite, hoarseness, and hair loss. Contact your doctor if these mild side effects persist or worsen.
Stop taking this medicine and contact your doctor immediately if you experience any serious side effects, including the following:
• Allergic reactions such as breathing difficulties, body itching, body rash, skin blistering, swelling of the face, mouth, tongue, lips, or throat;
• Anemia (low red blood cells) with symptoms like dizziness, pale skin, unusual tiredness;
• Blood clot in the arm or leg with symptoms like calf pain, chest pain, coughing up blood;
• Bone damage in the jaw with symptoms like mouth, teeth, or jaw pain; swelling or sores inside the mouth;
• Easy bruising or bleeding;
• Electrolyte imbalance with symptoms such as numbness and tingling in the hands, muscle weakness, fatigue, bone pain, fractures, and an irregular heartbeat.
• Fistula with symptoms like anal bleeding, pain with bowel movements, and fever;
• Heart problems with symptoms such as chest pain, cough, fast or irregular heartbeat, shortness of breath, weakness, or fatigue;
• Infection with symptoms like fever or chills, cough, shortness of breath, prolonged dizziness, headache;
• Liver problems with symptoms like nausea, vomiting, diarrhea, loss of appetite, weight loss, and yellowing of the skin or whites of the eyes;
• Neurological problems such as headache, seizures, weakness, confusion, high blood pressure, vision changes;
• Pancreatitis with symptoms like abdominal pain on the upper left side, back pain, nausea;
• Thyroid problems with symptoms like heat or cold intolerance, palpitations, hyperdefecation, or constipation

Cabometyx Precautions
Do not take this medication if you are allergic to cabozantinib, other ingredients in Cabometyx, or any other medicines. Before you start taking this medication, ensure that you inform your doctor about any prior or current medical conditions, including the following:
• Bleeding problems;
• Blood clot disorders;
• Electrolyte imbalances;
• Gastrointestinal problems such as peptic ulcers, intestinal perforation;
• Heart diseases;
• High blood pressure;
• Jawbone problems;
• Kidney disease;
• Liver disease;
• Stroke;
• Thyroid condition
Cabozantinib can cause drowsiness or dizziness, which may affect your ability to drive or operate machinery. You should refrain from driving, operating machinery, or performing other hazardous tasks until you know how it affects you. Combining grapefruit or grapefruit juice with cabozantinib may increase the risk of a buildup in the body and potential harm. Therefore, it is important to avoid consuming these products while taking this medication. Cabometyx can impair wound healing. If you have an upcoming surgery, including dental surgery, you will likely need to stop taking this medicine at least three weeks before the procedure. Please inform all your healthcare team about taking cabozantinib and discuss with your doctor if you need to discontinue it before surgery.
Avoid contact with people with measles, chickenpox, or the flu, as this medication can make you more susceptible to these infections. Taking cabozantinib during pregnancy could pose harmful risks to the developing fetus. It should not be used during pregnancy. If pregnancy occurs while on this medication, contact your doctor right away. Effective contraception is required during and for at least 4 months after treatment with this medication. It is also not recommended to breastfeed while using this medication.
Drugs like ketoconazole, clarithromycin, and ritonavir block the enzymes that clear Cabometyx. This causes the drug to build up to toxic levels, significantly increasing the risk of severe side effects like hemorrhaging or intestinal perforations. Therefore, before using this medication, inform your doctor about any other medications you are taking, including herbal medicines and supplements, to determine whether it is safe to use with cabozantinib. Take Cabometyx tablets orally on an empty stomach, either 1 hour before eating or 2 hours after eating. Swallow this medication whole; do not chew or crush the tablets. Avoid grapefruit-containing meals while taking this medication. Your doctor will determine the appropriate dosage for you. Use this medication as prescribed and avoid missing a dose. Store at room temperature and keep out of reach of children and pets.
