What is Galliprant (Grapiprant) Used For?

Galliprant (Grapiprant) is a prescription non-steroidal, non-cyclooxygenase inhibiting anti-inflammatory medication used for the treatment of pain and other symptoms of inflammation associated with mild to moderate osteoarthritis in dogs.

Its active ingredient, grapiprant, works by selectively blocking the EP4 receptor, which is a major receptor for a substance called prostaglandin E2. Prostaglandin E2 is the main substance responsible for causing pain and inflammation in dogs with osteoarthritis. When prostaglandin E2 binds to the EP4 receptor, it triggers several physical effects such as the widening of blood vessels and the production of more inflammatory mediators. Importantly, this receptor is the primary way that prostaglandin E2 makes sensory neurons more sensitive, which leads directly to the perception of pain. By specifically blocking this receptor, grapiprant prevents these signals from being activated, thereby reducing both pain and inflammation.

Older nonsteroidal anti-inflammatory drugs (like carprofen or meloxicam) work by blocking the COX enzymes that produce prostaglandin E2 in the first place. However, while this is effective in curtailing the pain and inflammation associated with prostaglandin E2, it does affect the beneficial effects of prostaglandin E2, such as protecting the stomach lining and maintaining healthy blood flow to the kidneys. This is ultimately why long-term use of these older nonsteroidal anti-inflammatory drugs carries a risk of stomach irritation and kidney strain. This risk is less associated with grapiprant due to its different approach.

What Side Effects Can Galliprant or Generic Grapiprant Cause?

The most frequently reported side effect is vomiting, which occurs in more than 1 out of every 10 dogs treated. Other possible side effects include loose stools, diarrhea, and loss of appetite. Consult your veterinarian if these side effects persist or worsen.

Several serious side effects occur very rarely, in fewer than 1 out of every 10,000 treated dogs. These include:
• Allergic reactions such as breathing difficulties, body rash, itching, hives, skin blistering, and swelling of the face;
• Bloody diarrhea;
• Vomiting blood;

Consult your veterinarian as soon as possible if you notice any of these serious side effects. Ensure you inform your doctor about any other side effects you notice that are not listed here.

Warnings

Do not use this medication in dogs with a history of allergic reaction to grapiprant, or any of the other ingredients used to make Galliprant tablets. Prostaglandins play critical roles in ovulation, implantation, fetal development, and uterine contractions. As a result, this medication is strictly contraindicated in breastfeeding and pregnant dogs or those intended for breeding. Also, do not start this medication in dogs that are actively dehydrated, anorexic, or experiencing vomiting or diarrhea.

Veterinarians must exercise caution when prescribing this medication to dogs with some conditions, especially the following:
• Existing gastrointestinal disease;
• Heart disease;
• Kidney disease;
• Liver disease;

Never give this medication at the same time as other nonsteroidal anti-inflammatory drugs. Combining them drastically raises the risk of severe stomach ulcers, intestinal perforation, or kidney failure. Other medications that may interact with galliprant include:
• Anticonvulsants, or behavioral drugs;
• Corticosteroids such as prednisone, prednisolone, dexamethasone, or triamcinolone;

Therefore, before giving this medication to your dog, inform your vet about any other medications your dog is currently taking, including herbal medicines and supplements, to determine if it is safe to use along with Galliprant. Observe a treatment-free period before starting Galliprant if you recently gave your dog a different anti-inflammatory product. This break helps you avoid potential side effects, though its length depends on how long the previous medication stays in your dog's system.

This medication has not been proven safe for puppies younger than 9 months old or dogs that weigh less than 3.6 kilograms. It is intended for use only in dogs. Do not use it in cats or other animals. Be sure to wash your hands after handling these tablets. Store this medication at room temperature and keep out of reach of children and pets to prevent accidental ingestion.
