Inlyta (Axitinib) Information

Inlyta, which contains the active ingredient axitinib, is a targeted therapy classified as a tyrosine kinase inhibitor (TKI). It is primarily used for the treatment of advanced renal cell carcinoma (kidney cancer) after one prior systemic therapy has failed. Inlyta is also frequently prescribed in combination with immunotherapy agents, such as pembrolizumab or avelumab, as a first-line treatment for patients with advanced disease. By inhibiting pathways that enable tumor growth and spread, Inlyta helps slow cancer progression. The medication is taken by mouth as a tablet, typically twice daily, approximately 12 hours apart.

The mechanism of action involves the potent and selective inhibition of vascular endothelial growth factor (VEGF) receptors 1, 2, and 3. These receptors are essential for angiogenesis—the process by which tumors create new blood vessels to supply themselves with nutrients and oxygen. By blocking these receptors, Inlyta "starves" the tumor of its blood supply, effectively inhibiting tumor growth and preventing the cancer from spreading to other parts of the body.

Inlyta Side Effects

Inlyta is generally well-tolerated, but some common side effects may occur as your body adjusts to the medication. Commonly reported effects include diarrhea, high blood pressure (hypertension), fatigue, decreased appetite, nausea, and a hoarse voice (dysphonia). Some patients may also experience "hand-foot syndrome," characterized by redness, swelling, or pain on the palms of the hands and soles of the feet.

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Blood Clots: Sudden pain, swelling, or warmth in an arm or leg, or sudden weakness on one side of the body.
• Gastrointestinal Perforation: Severe stomach pain, swelling of the abdomen, or persistent vomiting.
• Heart Failure or Heart Attack: Chest pain, sudden shortness of breath, or swelling in the ankles and feet.
• Severe Bleeding: Coughing up blood, dark or tarry stools, or unusual vaginal bleeding.
• Severe Hypertension: Symptoms of a sudden, dangerous rise in blood pressure, such as severe headache, confusion, or blurred vision.
• Thyroid Dysfunction: Unexplained weight gain or loss, feeling very cold or hot, or changes in heart rate.

Inlyta Precautions

Do not use Inlyta if you have a known allergy to axitinib or any other components of the tablet. Before starting treatment, inform your doctor of your complete medical history, especially if you have a history of high blood pressure, heart disease, liver disease, a history of blood clots, or recent major surgery. Because Inlyta can interfere with wound healing, you should typically stop taking it at least 24 hours before any scheduled surgery or dental procedure.

Tell your doctor about all medications you take, as Inlyta has significant drug interactions. Its levels in the blood can be significantly increased by strong CYP3A4 inhibitors, such as certain antifungals (e.g., itraconazole) and antibiotics (e.g., clarithromycin). Conversely, strong CYP3A4 inducers, such as the herbal supplement St. John's Wort or certain antiseizure medications, can reduce the effectiveness of Inlyta. You must avoid grapefruit and grapefruit juice, as they can interfere with how your body breaks down the medication. Your doctor should also monitor any blood pressure medications you are taking, as your dose may need to be adjusted while on Inlyta.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Inlyta can cause fetal harm and should not be used during pregnancy. Women of childbearing age should use effective contraception during treatment and for at least one week after the final dose. It is unknown if axitinib passes into breast milk; therefore, breastfeeding is not recommended during treatment and for at least two weeks after the last dose.

Take Inlyta exactly as prescribed, with or without food. Swallow the tablets whole with a full glass of water. If you vomit after taking a dose, do not take an extra one. Wait for your next scheduled dose. It is essential to monitor your blood pressure regularly at home while taking this medication. Store the tablets at room temperature in a dry place, away from heat and moisture.
