Kytril (Granisetron) Information

Kytril, which contains the active ingredient granisetron, is a potent 5-HT3 receptor antagonist. It is primarily used to prevent nausea and vomiting associated with initial and repeat courses of emetogenic cancer therapy, including high-dose chemotherapy and radiation therapy. It is also used to prevent and treat postoperative nausea and vomiting. Kytril is available in several forms, including oral tablets, an oral solution, and an intravenous injection, typically administered shortly before the start of medical treatment.

The mechanism of action involves blocking serotonin (5-hydroxytryptamine) at the 5-HT3 receptor sites located both on the vagus nerve terminals in the gastrointestinal tract and in the chemoreceptor trigger zone of the brain. During chemotherapy or radiation, mucosal enterochromaffin cells release serotonin, which normally stimulates these receptors, triggering the vomiting reflex. By selectively binding to these receptors, Kytril prevents the "vomit signal" from being sent, effectively suppressing the sensation of nausea and the urge to vomit.

Kytril Side Effects

Kytril is generally well-tolerated, though side effects can occur as the body processes the medication. Commonly reported effects include mild headache, constipation, diarrhea, fever, or a temporary feeling of weakness (asthenia). Some patients may also experience transient elevations in liver enzymes or a mild skin rash.

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Changes in Heart Rhythm: A feeling of a racing or pounding heart, or an irregular heartbeat (QT prolongation).
• Hypersensitivity Reactions: Sudden swelling of the face, lips, or tongue; hives; or difficulty breathing.
• Serotonin Syndrome: High fever, agitation, shivering, accelerated heart rate, muscle rigidity, or hallucinations (especially when taken with certain antidepressants).
• Severe Abdominal Issues: Severe constipation or abdominal pain/bloating, which could indicate a decrease in bowel motility.

Kytril Precautions

Do not use Kytril if you are allergic to granisetron or other 5-HT3 receptor antagonists (such as ondansetron/Zofran). Before starting treatment, inform your doctor if you have a history of heart rhythm disorders, electrolyte imbalances (such as low potassium or magnesium), or recent abdominal surgery, as the medication may mask signs of a bowel obstruction.

Tell your doctor about all medications you take. Kytril may interact with drugs that affect the heart's rhythm or other serotonergic drugs (like SSRIs or SNRIs), which can increase the risk of serotonin syndrome. Because Kytril can cause constipation, use with caution if you have underlying bowel motility issues.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Kytril should only be used during pregnancy if the potential benefit justifies the potential risk to the fetus. It is unknown whether granisetron is excreted in human milk. Therefore, caution should be exercised when administered to a nursing mother.

Take Kytril exactly as directed by your doctor. For chemotherapy, the oral dose is typically taken up to one hour before treatment begins. If you are taking the oral solution, use a calibrated measuring device to ensure the correct dose. Store the medication at room temperature, away from direct sunlight and moisture, and keep it out of reach of children.
