Manerix (Moclobemide) Information

Manerix (moclobemide) is a prescription medication used primarily to treat Major Depressive Disorder (MDD) and Social Anxiety Disorder (SAD) (Social Phobia) in adults. It belongs to a unique class of antidepressants known as Reversible Inhibitors of Monoamine Oxidase-A (RIMA). Manerix Side Effects

While Manerix is generally well-tolerated and often has fewer anticholinergic or weight-related side effects than older antidepressants like TCAs, some individuals may still experience adverse effects. The most common side effects reported are usually related to CNS stimulation and include insomnia (which can often be managed by avoiding evening doses), headache, dizziness, dry mouth, nausea, anxiety, and restlessness. • Hypertensive Reactions and Crisis: Although the risk is significantly lower than with irreversible MAOIs, consuming large amounts of tyramine-rich foods (e.g., aged cheeses, tap beer, cured meats) or mixing the drug with certain sympathomimetic medications can still trigger a rapid and dangerous increase in blood pressure (hypertensive crisis). Symptoms include a sudden, severe headache, confusion, blurred vision, and chest pain.
• Mania/Hypomania: Moclobemide can potentially precipitate a manic or hypomanic episode in patients with undiagnosed bipolar disorder, requiring immediate dose adjustment or discontinuation.

• Serotonin Syndrome: This is the single most critical risk and is a potentially fatal reaction that occurs when Manerix is combined with other serotonergic drugs (e.g., SSRIs, SNRIs, triptans, pethidine, or dextromethorphan). Symptoms may include agitation, hallucinations, a rapid heart rate, fever, muscle rigidity, and severe nausea. A mandatory washout period is required when switching medications.

• Suicidality and Worsening of Depression (Boxed Warning): Antidepressants, including Manerix, carry a strong risk warning for increasing the risk of suicidal thoughts and behaviour, particularly in young adults (under 25), especially during the initial phase of treatment or following a dose change.

Manerix Precautions

Before taking Manerix, it is essential to have a thorough discussion with a healthcare provider about your medical history and any other medications you are taking. Manerix is absolutely contraindicated for use with a vast number of different antidepressants, including all SSRIs, SNRIs, most TCAs, and certain strong analgesics (e.g., pethidine/meperidine) due to the severe, potentially fatal risk of Serotonin Syndrome; a mandatory washout period is required when switching between these medications. Caution is also required in patients with pre-existing conditions such as uncontrolled hypertension, severe liver impairment, hyperthyroidism, pheochromocytoma, or a history of seizure disorder or mania/hypomania.
