What is Mepron (Atovaquone) Used For?
Mepron (Atovaquone) is an antiprotozoal agent used for the
prevention of Pneumocystis jirovecii pneumonia (PCP) in adults and adolescents aged 13 years and older who cannot tolerate first-line treatment with trimethoprim-sulfamethoxazole. This serious fungal lung infection primarily affects individuals with weakened immune systems. It is also used to treat acute, mild-to-moderate Pneumocystis jirovecii pneumonia in adults and adolescents aged 13 years and older who cannot tolerate first-line treatment with trimethoprim-sulfamethoxazole. Furthermore, atovaquone (when combined with proguanil in fixed-dose combination tablets like Malarone) is used to treat and prevent malaria caused by Plasmodium falciparum.
Atovaquone works by disrupting cellular energy production and DNA synthesis in susceptible parasites and fungi. It binds to the cytochrome bc1 complex in the organism's mitochondria, thereby blocking electron transport. This action halts energy production, starving the pathogen. This electron blockade also shuts down dihydroorotate dehydrogenase, an enzyme essential for pyrimidine synthesis, preventing the pathogen from generating the raw materials it needs to reproduce or grow. Without the ability to generate energy or to construct its own genetic structures, the fungus or parasite is unable to grow or reproduce, effectively stopping the infection.

What Side Effects Can Mepron or Generic Atovaquone Cause?
Mepron is generally well-tolerated compared to other antiprotozoal medications, but it can produce a range of side effects depending on individual sensitivity and underlying health status. Common and mild side effects of this medication often involve the digestive tract, resulting in nausea, vomiting, diarrhea, and abdominal pain. Other possible side effects may include headaches, fever, fatigue, difficulty sleeping (insomnia), excessive sweating, coughing, a runny or stuffy nose, oral thrush, and a mild rash. Contact your doctor if these mild side effects persist or worsen.
Serious side effects may include:
• Allergic reactions with symptoms such as hives, skin blistering, swelling of the face, lips, tongue, or throat, or difficulty breathing or swallowing;
• Liver or kidney problems with symptoms such as yellowness of the skin or eyes (jaundice), severe abdominal pain, dark urine, unusual swelling, or changes in urination;
• Pancreatitis (inflammation of the pancreas) with symptoms such as sudden pain in the upper area of the abdomen (belly), nausea/vomiting, and fever;
It can also cause hematologic dysfunction, with rare reports of severe blood abnormalities (such as anemia, low white blood cell counts, low platelet counts, or methemoglobinemia).
Seek immediate medical attention if you experience any of these serious side effects. Inform your doctor about any other side effects you notice that are not listed here.
Warnings
Do not use this medication if you have an allergic reaction to atovaquone or any other ingredients in Mepron. Caution is advised when using this medication in individuals with some specific medical conditions such as:
• Diarrhea;
• Kidney problems;
• Liver disease;
Atovaquone levels can be significantly lowered by certain other medicines, specifically the antibiotics rifampin and rifabutin, so using these together is not recommended. Caution is also required when taking the antibiotic tetracycline or the stomach medication metoclopramide, as these can reduce the amount of atovaquone in the blood and potentially cause the treatment to fail. Therefore, before using atovaquone, it is important to inform your doctor about any other medications you are currently taking, including herbal medications and supplements, to determine if it is safe to use along with Mepron.
As there is currently not enough information from human studies to determine if this medication causes congenital disabilities in pregnancy and adverse effects in breastfed infants, consult your doctor before taking this medication during pregnancy or while breastfeeding to determine if it is safe for you and your baby. Also, the safety and effectiveness of this medication have not been established in pediatric patients under 13 years of age.
This medication is naturally difficult for the body to absorb and, as a result, must always be taken with a meal to ensure enough enters the bloodstream to be effective. Individuals with gastrointestinal disorders or those who have difficulty eating might not absorb a sufficient amount of this medication, and healthcare providers may need to consider alternative therapies for these individuals. Store this medicine at room temperature, between 15°C and 25°C (59°F to 77°F). Do not freeze. Keep out of reach of children and pets.
