Onglyza (Saxagliptin) Information

Onglyza, which contains the active ingredient saxagliptin, is an oral medication used to improve blood sugar control in adults with type 2 diabetes mellitus. It is prescribed when diet and exercise alone are insufficient and may be used alone or in combination with other antidiabetic medications, such as metformin, sulfonylureas, or insulin. Onglyza is taken by mouth once daily and is intended to support steady blood glucose control throughout the day.

The mechanism of action of saxagliptin involves inhibiting the enzyme dipeptidyl peptidase-4 (DPP-4). This enzyme usually breaks down incretin hormones, which are released after meals and help regulate blood sugar levels. By blocking DPP-4, saxagliptin increases incretin hormone levels, leading to increased insulin release when blood sugar is high and reduced hepatic glucose production. Because its effects are glucose-dependent, Onglyza has a low risk of causing hypoglycemia when used alone.

Onglyza Side Effects

Onglyza is generally well-tolerated, but some common side effects may occur. Commonly reported effects include headache, upper respiratory tract infections, sore throat, runny nose, nausea, and mild stomach discomfort. Some individuals may experience joint pain, fatigue, or mild swelling in the hands or ankles.

More serious side effects can occur and require immediate medical attention. If you experience any of the following symptoms, stop taking the medication and seek urgent care:
• Severe allergic reactions, including rash, hives, swelling of the face, lips, or throat, difficulty breathing
• Severe joint pain that may be sudden or persistent
• Signs of heart failure, including shortness of breath, rapid weight gain, swelling of the legs or ankles
• Signs of low blood sugar (more likely when used with insulin or sulfonylureas), including sweating, shaking, and confusion
• Signs of pancreatitis, including severe abdominal pain radiating to the back, nausea, and vomiting

Onglyza Precautions

Do not use Onglyza if you are allergic to saxagliptin or any ingredient in the drug. Before starting treatment, inform your doctor of your complete medical history. People with kidney disease, heart failure, a history of pancreatitis, or weakened immune systems may require dose adjustments or close monitoring. Your doctor will recommend assessing your kidney function before starting Onglyza and monitoring it periodically during treatment.

Tell your doctor about all medications you take, including prescription drugs, over-the-counter medicines, herbal supplements, and vitamins. Onglyza can interact with other diabetes medications, increasing the risk of hypoglycemia. Certain drugs that strongly affect liver enzymes (such as some antifungals or antibiotics) may alter saxagliptin levels, requiring dose adjustments. Alcohol should be used cautiously, as it may interfere with blood sugar control. Inform your doctor if you are pregnant or planning to become pregnant. There is limited information on the use of saxagliptin during pregnancy. This medicine should only be used if clearly needed. It is unknown if saxagliptin passes into breast milk. If you are breastfeeding, discuss potential risks and benefits with your doctor.

Take Onglyza exactly as prescribed, usually once daily, with or without food. Take it at the same time each day to maintain consistent blood levels. Do not stop taking Onglyza without consulting your doctor, as blood sugar levels may rise. Store the medication at room temperature, away from moisture and heat, and keep it out of reach of children.
