Scemblix (Asciminib hydrochloride) Information

Scemblix, which contains the active ingredient asciminib hydrochloride, is a targeted therapy medication used to treat adults with Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML). It is specifically used for patients who have already been treated with two or more other leukemia medications (tyrosine kinase inhibitors) or for those with a specific genetic mutation called T315I. The medication is taken by mouth as a tablet, typically once or twice daily depending on the specific condition being treated. Scemblix is taken by mouth as a tablet, typically once or twice daily, as prescribed by your doctor and depending on the specific condition being treated. The tablets should be swallowed whole with water; do not crush, break, or chew them. Scemblix can be taken with or without food, but it’s important to take your doses at the same times each day to maintain consistent levels in your body.

The mechanism of action of asciminib differs from that of older leukemia drugs. It is known as a STAMP inhibitor (Specifically Targeting the ABL Myristoyl Pocket). While other treatments bind to one part of the Bcr-Abl protein, Scemblix binds to a different, specific "pocket" on the protein to lock it in an inactive state. This blocks the signals that tell leukemia cells to multiply, helping to stop the progression of the disease.

Scemblix Side Effects

Scemblix is generally well-tolerated, but common side effects may occur as your body adjusts to the medication. Commonly reported effects include feeling very tired (fatigue), muscle or joint pain, headache, dizziness, and digestive issues like nausea, diarrhea, or constipation. Some patients may also experience cold-like symptoms, such as a sore throat or cough, as well as mild skin rashes.

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Blood cell changes: Fever, chills, or unusual bruising or bleeding may indicate low white blood cell or platelet counts.
• Heart or vascular problems: Chest pain, shortness of breath, sudden numbness or weakness, or swelling in the legs or arms.
• High blood pressure: Severe headaches, blurred vision, or pounding in your neck or ears.
• Pancreas problems: Severe pain in the upper stomach area that may spread to your back, often accompanied by nausea or vomiting.
• Severe allergic reactions: Swelling of the face, lips, or throat; hives; or significant difficulty breathing.
• Signs of fluid retention: Sudden weight gain or swelling in the hands, ankles, or feet.

Scemblix Precautions

Do not use Scemblix if you have a known allergy to asciminib hydrochloride or any of the inactive ingredients in the medication. Before starting treatment, inform your doctor of your complete medical history, especially if you have a history of pancreatitis, heart disease, high blood pressure, or high cholesterol (triglycerides).

Tell your doctor about all medications you take, as Scemblix has significant drug interactions. Scemblix can interact with a wide range of medications, including certain anticoagulants (blood thinners) and those metabolized by specific liver enzymes, which may increase the risk of side effects or reduce treatment effectiveness.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Scemblix can cause serious harm to a developing fetus and must not be used during pregnancy. Women who can become pregnant should have a pregnancy test before starting treatment and must use effective contraception during treatment and for at least one week after the final dose. Breastfeeding is not recommended while taking Scemblix and for one week after the last dose, as it could harm the nursing infant.

Take Scemblix exactly as your doctor prescribes. Swallow the tablets whole; do not crush, break, or chew them. If you miss a dose, check your schedule: if you take it once daily and are more than 12 hours late, skip the dose. If you take it twice daily and are more than 6 hours late, skip it. Never take a double dose to make up for a missed one. Store the tablets at room temperature in their original container, away from moisture and light, and keep them safely out of reach of children.
