Tagrisso (Osimertinib mesylate) Information

Tagrisso, which contains the active ingredient osimertinib mesylate, is a targeted therapy medication, a third-generation tyrosine kinase inhibitor (TKI). It is primarily used to treat adults with a specific type of lung cancer called non-small cell lung cancer (NSCLC). It is often prescribed when the cancer has spread to other parts of the body or as an additional treatment after the tumour has been surgically removed to help prevent the cancer from returning. Tagrisso specifically targets cancer cells with specific mutations in the epidermal growth factor receptor (EGFR). The medication is taken by mouth as a tablet, typically once daily. You can take it with or without food. To ensure the medication works effectively, take it at the same time every day. If you have difficulty swallowing tablets, your doctor may provide instructions on how to disperse the tablet in water.

The mechanism of action of osimertinib involves blocking the activity of the mutated EGFR protein. In certain lung cancers, this protein is overactive, signalling the cancer cells to grow and divide uncontrollably. By binding to these mutated receptors, Tagrisso blocks growth signals, helping shrink or slow tumour growth and spread. Because it is a third-generation TKI, it is specifically designed to target the mutations that often cause resistance to older lung cancer drugs.

Tagrisso Side Effects

Tagrisso is generally well-tolerated, but common side effects may occur as your body adjusts to the medication. Commonly reported effects include diarrhea, skin rash, dry skin, and changes in the fingernails or toenails (such as redness or tenderness). You may also experience feeling very tired (fatigue), mouth sores, or a decrease in appetite.

More serious side effects can occur and require immediate medical attention. Stop taking the medication and seek help if you experience:
• Blood clot signs: Sudden shortness of breath, chest pain, or swelling and pain in a leg or arm.
• Heart problems: Feeling like your heart is pounding or racing, shortness of breath, or swelling in your ankles and feet.
• Lung inflammation: New or worsening cough, fever, or difficulty breathing (which may indicate interstitial lung disease).
• Severe allergic reactions: Swelling of the face, lips, or throat; hives; or significant difficulty breathing.
• Severe skin reactions: A painful red or purplish rash that spreads and causes blistering or peeling.
• Vision changes: Eye pain, blurred vision, or sensitivity to light.

Tagrisso Precautions

Do not use Tagrisso if you are allergic to osimertinib or any of the ingredients in the tablet. It is important to inform your doctor if you have a history of lung or breathing problems, heart conditions (including Long QT syndrome), or electrolyte imbalances. Before starting treatment, disclose your complete medical history to your doctor.

Tell your doctor about all medications you are taking, including prescription drugs, over-the-counter medicines, vitamins, and herbal supplements (such as St. John’s wort). Tagrisso can interact with many other drugs, particularly certain antibiotics, anti-seizure medications, and heart rhythm regulators, which can change how the medicine works or increase the risk of side effects.

Inform your doctor if you are pregnant, planning to become pregnant, or breastfeeding. Tagrisso can cause serious harm to a developing fetus and must not be used during pregnancy. Women of childbearing age should use effective hormonal or barrier contraception during treatment and for at least six weeks after the final dose. Men with female partners who can become pregnant should use effective contraception during treatment and for four months after the final dose. Breastfeeding is not recommended while taking Tagrisso and for two weeks after the last dose, as it could harm the nursing infant.

Take Tagrisso exactly as your doctor tells you. Do not change your dose or stop taking it without first talking to your doctor. If you miss a dose, skip it and take your next dose at the regular time. Never take two doses at once to make up for a missed one. Store the tablets at room temperature, away from moisture and light, and keep them in a safe place out of the reach of children.
