Lynparza Information

Lynparza (Olaparib) is used for several specific cancer treatment scenarios, often requiring a companion diagnostic test to detect specific genetic markers. Lynparza is typically used as a maintenance therapy for the treatment of cancer of the ovaries, fallopian tube, or primary peritoneal cancer and can be taken alone or in combination with bevacizumab. It is administered after the initial treatment phase has ended, to prevent the progression of the cancer or keep the cancer from coming back. In conjunction with bevacizumab, it is used if your advanced ovarian cancer has tested positive for homologous recombination deficiency and has responded to your first treatment with platinum-based chemotherapy. It may also be used alone in advanced ovarian cancer that has tested positive for a BRCA mutation and has responded to initial treatment with platinum-based chemotherapy.

It is also used in the treatment of breast cancer, specifically for HER2-negative patients with a germline BRCA mutation. It is used in early-stage breast cancers with a high risk of recurrence, where it is given after surgery. You should have received chemotherapy medicines before or after surgery to remove the tumor. It is also used in advanced breast cancer that has spread to other parts of the body (metastatic). Following the diagnosis, you would have received chemotherapy before or after the cancer spread. If your cancer is hormone receptor-positive, you should have already undergone hormone therapy, or you are not a good candidate for that treatment. Lynparza can also treat pancreatic cancer when it has spread (adenocarcinoma) and is associated with a specific BRCA gene mutation. It's usually used as maintenance treatment after a patient's cancer has not progressed during at least their first 16 weeks of platinum-based chemotherapy.

Furthermore, Lynparza is beneficial in treating metastatic castration-resistant prostate cancer driven by specific genetic mutations in the homologous recombination repair pathway. It's used when the cancer has metastasized and continues to progress despite standard treatments like hormonal therapy (lowering testosterone) or other cancer medicines known as enzalutamide or abiraterone. Lynparza is also approved for the treatment of advanced prostate cancers with specific mutations in the BRCA genes. When cancer spreads to other parts of the body and no longer responds to conventional treatment, including testosterone suppression, Lynparza is often combined with abiraterone and prednisone or prednisolone.

Lynparza (olaparib) is a fascinating drug, particularly because it represents a shift toward more targeted cancer treatments. It contains the active ingredient olaparib, a potent PARP (poly ADP-ribose polymerase) inhibitor. Olaparib inhibits the PARP enzyme, which repairs single-strand DNA breaks. While healthy cells typically have functional backup repair pathways (like homologous recombination), cancer cells with BRCA mutations lack these alternatives. By inhibiting PARP, olaparib forces these cancer cells to accumulate catastrophic genomic damage that they cannot fix, ultimately triggering programmed cell death (apoptosis). This targeted approach appears highly effective at preventing disease progression after initial chemotherapy, as it specifically exploits the tumor's preexisting genetic weakness while sparing a significant portion of healthy tissue. Consequently, it extends progression-free survival and, in some clinical settings, has shown a meaningful impact on overall survival in patients with high-risk genetic profiles.

Lynparza Side Effects

Like other chemotherapies, olaparib can cause some side effects. Common mild side effects of this medication include a change in taste, constipation, cough, diarrhea, dizziness, headache, heartburn, loss of appetite, mouth sores, nausea, rash, tiredness, vomiting, and weakness. Contact your doctor if these mild side effects persist or worsen.

Stop taking this medicine and contact your doctor immediately if you experience any serious side effects, including the following:
• Allergic reactions such as breathing difficulties, body itching, body rash, skin blistering, swelling of the face, mouth, tongue, lips, or throat;
• Blood clot problems with symptoms like pain or swelling in an extremity, shortness of breath, chest pain, breathing that is more rapid than normal (tachypnea), or heart beating faster than normal (tachycardia);
• Bone marrow problems called myelodysplastic syndrome or acute myeloid leukemia with symptoms such as weakness, weight loss, fever, frequent infections, blood in urine or stool, shortness of breath, feeling very tired, bruising or bleeding more easily;
• Liver injury with symptoms like discomfort on the right side of your stomach area (abdominal), dark or tea-colored urine, or yellowing of your skin or the whites of your eyes (jaundice);
• Lung problems (pneumonitis) with symptoms like new or worsening symptoms of lung problems, including shortness of breath, fever, cough, or wheezing

Lynparza Precautions

Do not take this medication if you are allergic to olaparib, other ingredients in Lynparza, or any other medicines. Before you start taking this medication, ensure that you inform your doctor about any prior or current medical conditions, including the following:
• Active infection;
• Blood count disorders such as anemia;
• Blood clot problems;
• Breathing or lung problems;
• Kidney disease;
• Liver disease

Your doctor will do blood tests to check your blood cell counts and other parameters before and after starting this medication to ensure it is suitable for you and to monitor for side effects. Before you do tasks that may require special attention, wait until you know how you respond to Lynparza. If you feel dizzy, weak, or tired, do not drive or use tools or machines. Pregnancy should be avoided while taking olaparib due to potential risks to the developing baby. If you or your partner becomes pregnant while taking this medication, contact your doctor right away. Women of reproductive age group taking this medication should use effective contraception for at least six months after the last dose. Men taking this medication are advised to use effective contraception for at least three months following the last dose. Do not breastfeed while taking this medication. It is recommended to avoid breastfeeding until at least one month after the last dose of the medication.

Medications like certain antifungals (ketoconazole, fluconazole), certain antibiotics (clarithromycin), and some heart medications (diltiazem, verapamil) can cause olaparib levels to spike, increasing the risk of toxic side effects. Therefore, before using this medication, inform your doctor about any other medications you are taking, including herbal medicines and supplements, to determine whether it is safe to use with Lynparza. You’ll need to avoid grapefruit and grapefruit juice as these act as inhibitors and can unpredictably and dangerously raise the concentration of this drug in your blood. Take Lynparza tablets orally as prescribed by your doctor or as stated in the medication pack. The usual dose is 300 mg, split into two 150 mg doses taken twice a day. Swallow the tablets whole with a glass of water, ideally at the same time each day. Do not chew, crush, or break the tablets. It’s important to follow the prescribed dosage and schedule, as your doctor may adjust it based on your specific condition. Store this medication at room temperature, and protect it from light and moisture. Keep it out of the reach of children.
